CADENCE: Evolutionary Fragment-Based Discovery of Novel ABL1, HER1,

Record of the development of the deep learning pipeline CADENCE, capable of generating novel competitive kinase inhibitors from amino acid sequence. CADENCE was trained on the Davis dataset of kinase inhibitors and binding scores, creating a binding affinity prediction model. This model was integrated into a feedback loop that utilized random sampling from the Davis dataset, diversity-aware selection, and Tanimoto similarity to generate and select candidates.
